Okay guys, this is just but a theory, so when you're done reading this just place your thoughts below,
and anything that i might of have left out.
So many people say that he could've been assasinated, others think maybe he got poisoned.
And when people read this page, they thought it was suicide.
But it didn't happen.
Now for the explanation on how he died, i was reading around the wiki a bit and i came up with this.
This is Tsunade's technique, ''Creation Rebirth''
(Text directly from the wiki)
''by releasing the great volume of chakra stored in her forehead at once, the body's cell division is forcibly stimulated by proteins, reconstructing all organs and all tissues making up the human body. The technique itself does not regenerate the old cells, rather it hastens the creation of new ones through division.''

''However, a body's cells can only split a certain number of times in a lifetime, and by speeding up this process, Tsunade is basically shortening her natural lifespan.''
Okay, now that that's done, take a look at this page.
After Tsunade gets stabbed in the stomach by Madara's susanoo sword, it regenerated the same second.
Afterwards he says that it it's the same ability Hashirama had.
And ofcourse with Hashirama growing up in the time of war, there must've been times where he needed to use it.
Having that said, it is same to presume (atleast for me it is) that he used this many times, and died young eventually.
