Based on that scan of Tsunade, it seems possible that she could have identified each Pain as a corpse, which might have been enough of a clue for Jiraiya to realize that the real Nagato was the one controlling them, which would have prompted them to search him out and kill him, leading to the defeat Nagato knew was possible if his secret was discovered.

By secret I guess he meant about the abilities of the 6 Peins. The only reason Jiraiya lost was due to him being unable to take into account the Naraka path's ability to repair the defeated Peins. Without that he probably would've won by himself.
Anyway Tsunade couldn't tell that the Peins are corpses even when Deva Pein appeared before her. So I doubt it'd would've been that simple.

Well based on the fact that in Part I a rusty Tsunade could feel Orochimaru's heartbeat, clearly Tsunade was nerfed in Part II, as she would have easily been able to tell that 1. They don't have a heartbeat, 2. Their skin is ice cold, and 3. Their complexion is that of a corpse.

But in the end it doesn't matter, and Pain's secret could have been either of those things.

The Pain bodies were just animated corpses, Naruto biology is definitely iffy but their inside organs aren't functional otherwise Hanzo's poison would've affected them.

Personally, Tsunade could just stick a Katsuyu on Jiraiya and her intelligence would just prove the upper-hand. Katsuyu will continuously heal Jiraiya's injuries and the Pein's would get destroyed 1 by 1.

This would be an amazing fight. I think Pain would win High Diff, but it's hard to say since we never see Jiraiya have to fight Tendo. In the end, they would lose for two reasons. Raw power of tendo, and lack of intel on the 6 paths.
