It is indeed because of its purity, the power of the fruit is too hazardous for humans without mitigation. If it is eaten, this purity becomes akin to a curse and the consumer steadily becomes extremely abnormal and added to this abnormality, it causes the consumer’s mind to deteriorate and this deterioration of the mind causes the body to complete neglect purpose and pursue only Chakra. ]. Whether the Shinju itself made such a thing subtly known to mankind while executing it's creative ability or whether it was deducted is uncertain. The power of the Shinju, its fruit, gradually mutated Kaguya Ōtsutsuki into the Jūbi by its natural composition: the chakra that resides in the fruit is an untainted mass of energy. It is chakra as it was originally made to be, perfect.
It cannot be said for certain, but perhaps this perfect energy, this chakra, is too much for a human body to bear without a form of attenuation. A perfect chakra, such as the Shinju’s fruit could perhaps hypothetically damage to body in a similar fashion to how the Upper-Gate levels damage the body; however, instead of destroying the body like the Gates do, the Shinju’s fruit, its chakra, sturdily, and unceasingly ‘amplify’ the consumer’s anatomy causing abnormalities. This amplification would happen continually until it would periodically have an excess amount of chakra that needs to protrude from the individual’s body; and the abnormalities would range from genetic alterations to mental instability, which would have the individual, the consumer, needlessly craving more of the same chakra or similar chakra until he or she is content.

In essence, the changes Hagoromo mentioned taking place within individual's who gain or are seeking to gain omnipotence are easily seen. Madara has not only had negative alteration's mentally, that is, the instability and lapses in his rational and behavior currently but there are also anatomical changes. Hagoromo notes that Kaguya suffered identical effect's which brings about the plausibility----sides her absence to face the Jubi along side her sons. The sin of Kaguya was usurping the fruit and the end result of that or in context 'the consequence she paid' was a rampaging Jubi.
Kaguya may be the Jūbi.
Gratitude must be given to Varrah for his supernal thought's on the matter as without them the above theory would not be possible.
