Lets say there are two brothers from the Uchiha clan. They both have Mangekyō Sharingan. If they switched eyes with each other would they get EMS?

Well, this could work only if the brothers are not extremely close. Since acquiring the Mangekyou requires one to kill their best friend, a close set of sibling such as Madara&Izuna or Sasuke&Itachi could not achieve this. However, if the siblings are closer to, say, a best friend or distant-ish relative, this could possibly work.

I think I read something from Itachi that many brothers tried to attain EMS by killing their brothers and taking their eyes, but failed there might be some other prerequisite.
