Deidara didn't mean it like that, it was kinda like a figure of speech, per se. I doubt anyone in the Akatsuki knew about Itachi's love for Sasuke. If anyone, it'd be Kisame, not Deidara.

However, there are LOTS of early hints that Itachi cared for Sasuke in the beginning. You can tell from his body language towards him. Like in the hotel, when they first met, he didn't just drop him on the ground, he carefully placed him against the wall (albeit after the Tsukiyomi), etc etc. That's why everyone assumed that Itachi cared for Sasuke, because there were so many hints.

My favourite is (even though it's a bit later in pt 2) when Itachi said, "Did you get a little taller?"
