Don't know what website you got that page from, but in most Madara says he will not summon the PS again (disgraceful).
During the fight with Madara's Perfect Susanoo, was Tsunade mysterious handseal..

hmm.. true, but i don't know how much chakra Tsunade has currently.. maybe Sakura is there to fill out the rest of chakra needed to summon the 1/10 Katsuyu.. though, honestly, i don't know how much chakra is needed to summon Katsuyu in her entirety, though it maybe gargantuan.. though a very weakened and near death Tsunade still had enough chakra to summon a mid size version of Katsuyu's standardized size..

Well logically a rested Tsunade should have enough chakra to summon at least 5/10 of Katsuyu's original body. But probably she can't release so much chakra in a moment or something like this.

It seems like - Tsunade was trying to release more chakra to prepare her moves to fight against Madara. Just a guess.
But it doesn't look like preparation for summoning.

after knowing that Katsuyu is a damn huge thing,
I also thought that the hand seal she performed back then was to summon her in a bigger version to counter the PS
