Wins.

The intricate ability to continuously harness Shizen Enerūgi; that is, Jūgo's ability is one that can be at the Ryūchi Cave in conjunction with Sage Mode and is thereby applicable to Itachi. Zetsu highlighted that Itachi's performance while facing Sasuke was a far cry from what he usually capable of, specifically: reflexes, continuity, output and approach. Though this may have been done intentionally or unintentionally, Slaeht, I was hoping that you would have given a mindset for this match as Sasuke is Itachi's brother and realistically his approach to facing him could vary. My argument is based on Killing intent however; if this is not the case , then I'll make the necessary adjustments. Itachi having Sage Mode basically means he is able to stabilize Natural energy with his own and distribute it's properties across his jutsu and Sharingan as you've already stated. It means that Itachi's ocular abilities [from basic to specific] would all be amplified. Susanoō would become a Senjutsu Susanoō, Amatearsu's volume, head and execution speed would increase exponentially; Lastly, Itachi's Ninjutsu, Genjutsu, and Taijutsu all become's substantially more powerful. Sasuke's array of Enton particularies are pretty much useless as the Sharingan's perceptive ability would enable Itachi to react accordingly as well as the force of Nature being an insurance for evading incoming attacks. Susanoō [all types] sword swipes are defended against by Itachi. Sasuke cannot out run Itachi, nor entirely evade him given his enhanced Sharingan and Shun-Shin feats which are now being powered by Senjutsu.

Conclusion.

In the end as both Inton and Yōtōn compose it: The Totsuka no Tsurugi can seal Susanoō. Itachi stabs and seals Sasuke's Susanoō seals him along with Sasuke himself. Anything Sasuke does during this time is negated by the Yata Mirror.

The Sword of Totsuka can seal Susanoō?

Go on....i'm interested.

When both Itachi and Sasuke were affected by Hakugeki, Sasuke was completely disabled and about to die. Itachi, on the other hand, was able to move through the pain of the technique and summon Susano'o and protect Sasuke. If he can do that, Itachi can still use Hakugeki to disable Sasuke and follow it up with Muki Tensei, even if he's in exeruciating pain.

Perfect Susanoō isn't bypassing Yata-no-Kagami nor would it keep Itachi away from Sasuke, Strict. This is a healthy Itachi with high chakra reserves, Sennin Mōdo and Jūgo's ability.
As it is EMS Sasuke, who doesn't suffer from Chakra loss due to the usage of his MS techniques. Though Itachi's Susanoo possesses Totsuka and Yata, he can't overcome the power of the PS. Totsuka certainly can't seal Sasuke's Susanoo. It seals away one's soul, Susanoo is the mere manifestation of Chakra. Nagato, despite being an Edo, had his soul bound to a host. Against anything else, Totsuka works like a mere offensive weapon.

Genjutsu aren't working on either of them, Amaterasu can't affect Sasuke due to Enton, while it would still affect Itachi.

So the question is what is stronger, Itachi's Susanoo or the PS of Sasuke.

As far as we've sen so far, it seems EMS users are descendants of Indra. That means only descendants of ashura can compete. In other words, No shinobi, no sage, no nothing except a descendant of Ashura/Juubi jin is taking on an EMS user. PS is simply too much.

Nahh Strict, I'll agree with you on all points besides your conclusion [that it only boils down to who has the more powerful Susanoō], Consumption arising from using the Mangekyō Sharingan being a short term issue for Itachi as he was able to keep the light of his Mangekyō Sharingan for years and that he was also able to reduce this consumption on Chakra and stamina to the point that he could maintain a activate a complete Susanoō then proceed to seal Orochimaru and negate paper bombs sometime after; all this being down while Itachi was low on Chakra reserves to the point that he couldn't maintain a Three Tomoe Sharingan and was being impacted heavily by his sickness. It only point's out that this Healthy Itachi with high chakra reserves would be able to battle for extended periods, it's not going to be a short term issue.

Lastly, the bold as you're implying that Totsuka only seals souls which isn't the case. The databook states that the katana is saturated with sealing jutsu but the souls which are sealed are placed under Genjutsu for eternity and likewise sealed away. Orochimaru's Yamata no Jutsu was also a manifestation of his Chakra yet it was sealed within the sword and also as you rightly stated, Nagato's soul was attached to a host; just as Orochimaru's Yamata. Neither the 'dead host' or the 'Hydra' possessed souls yet they were sealed. The same concept it applicable to Susano'o. If Itachi plunges the blade through Sasuke's Susanoō it will seal it.

Totsuka-no-Tsurugi's blade is imbued with with sealing jutsu in that; seals anything it pierces, that is, both and things however, souls that are sealed within the gourd are placed under Genjutsu. Susanoō is created from Inton and gains a form through Yōton [the physical energy taken from the user]. The Totsuka no Tsurugi can seal Susanoō as it is really a tangible manifestation of the user's chakra.

Your logic is futile. If the sword seals everything it pierces, where do those inanimate objects go? Inside the gourd? No, because it seals only souls therefore they cannot be sealed as they don't have any place to go. Yōton and Inton are not what define a soul, they are merely an mimic practice; Inton is used to create a form and Yōton is what breathes life. But they still don't have a soul. What Nagato had was a soul, undoubtedly. That's why he could be sealed, while inanimate objects that don't have a soul cannot.

What about BM Minato,8th gate guy, Nagato, Juubito??? Rinnegan Obito,??, id say all of these can eat EMS Sasuke if we go by feats

They all die. Ems sasuke as PS no matter how desperately people try to deny it. Based on the fact that madara's ps = indras ps in size, we know sasuke's own will also be that big seeing as he's a reincarnation with the same powerful chakra.
The very susano'o he wrapped around bm naruto is ps and it had enough firepower to shatter through omnyoudon shield. That's enough.

Edit: i said juubi jins would also win lol. They're tapping into part of the power of sage, who's stronger than ashura and indra

Didn't say: "Yōton and Inton define a soul"...

Yet you're begging the question as what you conclude is only true in context of a false premise, that: "Totsuka-no-Tsurugi only seals souls"; something which was never directly stated or implied and contrarily to that the opposite is what has been portrayed throughout the manga. If "Totsuka-no-Tsurugi only sealed 'souls' then it would have sucked Nagato's soul out of the dead corpse, leaving it behind as it has 'no place to go'----applicable to Orochimaru's Yamata as well.
