An argument can be made: the Eternal Mangekyō Sharingan allows for more leisure usage of the otherwise strenuous abilities rose from the Mangekyō Sharingan, enabling Sasuke a pretext for defeating Itachi.​

Replacing Itachi's Sharingan with Senjutsu?

I am certain I did not imply this; but if I did, the answer is no: rather than Itachi and his Sharingan being replaced by Senjutsu, Senjutsu may be utilized alongside Itachi and his Sharingan.​

Itachi stands little chance of winning this.

-Hakugeki is useless without Suigetsu's abilities since he won't be able to move while its active.

-Muki Tensei is useless as it couldn't even penetrate Ribcage Susanoo.

The only hope is that Senjutsu will buff his Susanoo up to a level where he can bust through Sasuke's Armored Susanoo. Even then, Sasuke has PS, and no Senjutsu boost will help him then.

at the Ryūchi Cave in conjunction with Sage Mode and is thereby applicable to Itachi. Zetsu highlighted that Itachi's performance while facing Sasuke was a far cry from what he usually capable of, specifically: reflexes, continuity, output and approach. Though this may have been done intentionally or unintentionally, Slaeht, I was hoping that you would have given a mindset for this match as Sasuke is Itachi's brother and realistically his approach to facing him could vary. My argument is based on Killing intent however; if this is not the case , then I'll make the necessary adjustments. Itachi having Sage Mode basically means he is able to stabilize Natural energy with his own and distribute it's properties across his jutsu and Sharingan as you've already stated. It means that Itachi's ocular abilities [from basic to specific] would all be amplified. Susanoō would become a Senjutsu Susanoō, Amatearsu's volume, head and execution speed would increase exponentially; Lastly, Itachi's Ninjutsu, Genjutsu, and Taijutsu all become's substantially more powerful. Sasuke's array of Enton particularies are pretty much useless as the Sharingan's perceptive ability would enable Itachi to react accordingly as well as the force of Nature being an insurance for evading incoming attacks. Susanoō [all types] sword swipes are defended against by Itachi. Sasuke cannot out run Itachi, nor entirely evade him given his enhanced Sharingan and Shun-Shin feats which are now being powered by Senjutsu.

Conclusion.​

In the end as both Inton and Yōtōn compose it: The Totsuka no Tsurugi can seal Susanoō. Itachi stabs and seals Sasuke's Susanoō seals him along with Sasuke himself. Anything Sasuke does during this time is negated by the Yata Mirror.

Sasuke's Susano'O can withstand at least as much as Itachi's, while he has much better offense. Take in the factor Rinnegan also, that grants access to Mazō. The soul dragons are intangible, so Yata no Kagami wouldn't be able to negate, intercept nor block them at all.​

If Itachi were to be given Sage-Mode, we must automatically assume his chakra reserves are enormous. Logically speaking, he should have superior stamina to EMS Sasuke if that is the case. As others pointed out, giving Itachi such a Sage-Mode along with his Ocular powers would already make him a vast match for Sasuke, regardless of the restrictions. I think this battle would be prolonged, and Itachi would result as winner due to superior stamina.

things however, souls that are sealed within the gourd are placed under Genjutsu. Susanoō is created from Inton and gains a form through Yōton [the physical energy taken from the user]. The Totsuka no Tsurugi can seal Susanoō as it is really a tangible manifestation of the user's chakra. ​

Perfect Susanoō isn't bypassing Yata-no-Kagami nor would it keep Itachi away from Sasuke, Strict. This is a healthy Itachi with high chakra reserves, Sennin Mōdo and Jūgo's ability.​
