I believe that somewhere Sasuke said that he was going to kill Tobi. I don't remember when/where he said this but I'm fairly certain he said it or thought it ( but don't quote me on this). Not to mention he has seen Tobi's face before in the cave after the fight with Itachi, so he knew what he looked like. Also Madara is currently on the battlefield... RIGHT NOW, it would be quite obvious to tell that Tobi is not Madara.

Sasuke easily realized that Tobi wasn't Madara since Madara is currently on the battlefield, but by now he does know that Tobi.

Also it is shown right here he knows madara is edo'd before hes even on the battlefield

I can conclude that he does know that Obito is Tobi by face recognition from when he saw his face in the cave. He also knows that Tobi isn't Madara since Madara is on the battlefield, and that he knows that Tobi is Obito because Minato has said so right next to Sasuke.
