Hashirama was the first one to sense it.. He later called Tobirama dull for not sensing it from the start.

He saw the Rinnegan. If Hashirama had knowledge on the Rinnegan's forbidden jutsu "Rinne Tensei", that means its likely that he knows about the fundamentals and prerequisites in regards to obtaining the Dojutsu.

It is known to Hashi that Madara possesses Rinnengan and to get rinnengan you need Senju DNA. On some level Hashirama must know Madara at least has some Mokuton jutsu
