Do the Hyuga clan know who Tobi is?

I think they do because of their byakugan. It can see through many things and Tobi's mask is one of the many things it can see through. The Hyuga clan is just keeping his identity on the low for their sake of living. Won't be underestimating the byakugan again, will we? U_U

No, they don't. The Byakugan can see chakra networks in a shinobi's body, but they can't see the actual face. So they don't know how Tobi is. Maybe if someone older from the clan could see his chakra network and recognize him from when he wasn't wearing a mask, but somehow I doubt there is such a person in the Hyuuga clan.
Plus even if they could see his face, it still wouldn't work, because Tobi himself has stated his mask is made from something special.
