Shisui is the too nice type of guy so I don't think he would have resorted to killing people.

He might have awakened his MS probably at the death of his father, or since the Leaf Village was going through turmoil at this time he might have gotten it through skill. You don't need to awaken the MS just because you are butthurt and are lusting revenge. There are always other reasons.
