This he was there when Gai told them about Primary Lotus and Lee was the only one who could learn it, iirc primary lotus opens the first gate so he and tenten both knew Lee had learned about opening gates, it's unclear whether they knew how many levels he could unlock. I imagine they did.

That scan literally shows that he did know and he believed it wouldn't matter Lee would still lose, not only did he know about them he underrated them as well.

I'll sum it up for you since you didn't seem to understand it, Neji said:

Neji knew what the Gates did to the body and he knew Lee could and would use them, in Neji's opinion though this was a power not accepted by fate so he saw them as trivial at best.
