It's simply a byproduct of having Indra and ashuras chakra in a sharingan.

That gave him EMS. Which you need before the Sharingan. He got the Ashura DNA from Kabuto who supplied him with Hashirama's Senju cells when he was healing him from being stabbed by Madara.

If itachi were to awaken Rinnegan he would need

1: Sasuke (his brother's eyes) for EMS and as a bonus those eyes have indra chakra in them
2: DNA from Either Naruto or Hashirama (Ashura reincarnates)

Then he would awaken the Rinnegan.

Then heaven help the NV if Itachi were to succeed in this.

Nobody in the manga has ever said senju DNA.
He said hashiramas cells and chakra.
And it is common sense now that hashiramas cells effects were a result of the ashura chakra, he doesn't have magic cells.
