Obviously the next step before he could attain the Rinnegan would be the Eternal Mangekyou Sharingan. If he had defeated Sasuke, he would have taken his eyes and got an EMS. And since he has Senju DNA, that unlocks the Rinnegan as well.

The eye itself is DNA. Just because we've only seen Uchihas awakening it by getting Senju DNA doesn't mean it can't work the other way around (a Senju stealing an Uchiha's eye) or what Danzo did (Getting Hashi DNA implanted in him + implanting Shisui's eye + 10 sharingan)

One does not awaken the Rinnegan simply by mixing Senju and Uchiha DNA. That's something that can only be done by Indra and Ashura's reincarnations. Indra and Ashura's chakra =/= Senju and Uchiha random clan members chakra.
Who says some random pair of MS eyes allows one to awaken the MS? So far that only worked between brothers.
Even if that method worked, Danzo would certainly die before he awakened it (considering Madara had to wait since being a young adult until being a very old man to obtain it, while Danzo was already an old man when he fought Sasuke).

No eye supplies enough or any DNA of another person.

Kekkai genkai goes far beyond the eyes, you need the blood of an uchiha
