Can people like Sakura or Tsunade handle the Rinnegan?

No. Rinnegan eats away at the user's life force. As much as Byakugou replenishes the user from external injuries and in some instances internal injuries (depending on proficiency) life force can't be somehow restored through Medical Ninjutsu

Hence why Madara bestowed it to an Uzumaki, and they are known to have an incredible life force

Don't put Tsunade and Sakura in the same bucket. One is a direct descendant of Senju/Uzumaki and the other nobody cares. I am sure Tsuande could handle it.
But I think if Sasuke or Naruto ever die then Sakura could use it to sacrifice herself to bring them back. But I doubt she'd be able to do even that.
