So in chapters 594-596ish and episodes 511/512 we get reactions of a bunch of people to Luffy returning to Marineford.
One of these reactions is Buggy. In the manga all we get is him returning to his original crew and telling Alvida about John's treasure mark before Mr. 3 interrupts with the WG news.
In the anime in between John's treqsure and the WG news, Alvida shows him the news report about Luffy. Buggy's reaction is annoyance Rayleigh is with Luffy then wonders why he is with Luffy.
Here's the detail. Buggy starts thinking about Luffy's family "If I remember right his father is Dragon. His grandfather is Garp.....Rayleigh could be his uncle!!!!"
Now you could write this off as a sime.gag as it is Buggy doing it but on the other side of that Buggy does know Rayleigh pretty well and there may be truth to it.
Rayliegh's sister married Dragon? Maybe Buggy heard stories from Rqyleigh about his sister and her dangerous husband and is now putting two and two together.
