Me thinks the 2nd knows/suspects how Madara survived the fight with Hashirama. I highly doubt it was as simple as injecting Senju cells into him, as Kabuto wouldn't need anyones advice to do that...

But he says "using my knowledge and someone's advice" which I thought sort of implied it was advice to do with saving sasuke. But yeah your interpretation could be right too.

I posted a threat , talking about this one earlier today , that caught my attention too !! but it seems people mostly believe he was talking about Tobirama !! I personally doesn't have any guess about it !

This someone could be Tobirama (the nearest character with high knowledge for saving Sasuke), or Itachi (by the advice that moved Kabuto towards this answer of saving Sasuke)
