Nope it doesn't, hand signs aren't required to use the summoning jutsu. It's an optional thing, there are plenty times in the manga where you see summoning jutsu without hadpnd signs. Minato summoned Gerotora without hand signs, Naruto summoned Gamakichi without hand signs. All that is required is blood.

Besides we've already seen minato use hand signs with just 1 hand.
