Kaguya's Ōtsutsuki isn't quite the same as Hamura's Ōtsutsuki offspring. The latter's is a sort of neo-Ōtsutsuki Clan and although related, not the same clans; they just inherited their ancestral clan's name.

After Hamura's generation, that's how things went.

To awaken the Tenseigan, one needs the chakra of a main house Ōtsutsuki and the Byakugan of a main house Hyuga. This is because they are considered the pure, "undiluted" descendants of Hamura.
