Well nobody has a chakra like Hashirama so they can't use it the way he does. However I think Obito usage wasn't because of chakra conservation but because of his fighting style or plot U_U Obito constantly uses his Mangekyo, He controlled Gedo Mazo and tailed beasts over a large period of time. He's done so many chakra tanking things for long periods, he must have massive reserves U_U

Only Hashimara can use Mokuton.
If this Mokuton was for pure Bloodline Senju - SO6P younger son.
Using DNA may not be possible.

It was Kabuto that added somethings extra "Special version" to Madara body.
