The manga made it clear (I can dig up chapters if needed) one can sign the scroll of more than one animal. As far as we know, only Toad users have attained Sage Mode. The thought struck me, ninja can attain Sage Mode with any animal, they just have not spent the time with the animal to do so...

Itachi is the stronger of the two at this point. When they fight, Itachi still is simply playing with Sasuke, pushing him. Itachi is sickly and dying, but still has the ability to do so.

Zetsu, in my opinion, is the weakest. His only proven tech's were to multiply and look like other stuff. Everyone else has some other combat ability which greatly helps them.
