Unfortunately from what I have to come to see in the series, it is unlikely that a standard substitution would work in escaping the clutches of the black flame unless one had used it before the flames even formed, however advanced substitutions such as Orochimaru substitution style and Bee's sacrificing of one of Hachibi's tails have proven to outdo the mighty Amaterasu.

no. Sub works by switching your body with the sub BEFORE you actually get hit and making it look like you. It wouldn't work if you got hit by Ama. You could make it seem like you got hit though, but it was actually your sub.

No, substitution works BEFORE Amaterasu hits, not during or after.

Sub is like switching with a transformation. That is why Sakura did not actually take damage from the attack, she wasn't actually hit.
