There is a theme, but it's imperfect.
For the context of classification, a Rikudō is anybody that combined the two peaks of the two greatest bloodlines in the world. That is the peak of Senju, and the peak of Uchiha, ability. This means those ninja with both a Mangekyō Sharingan, as an acceptable peak of Uchiha ability, and poweful Senju [Hashirama or possibly Uzamaki] cells. The only ninja to do this perfectly is Madara, who possessed two Eternal Mangekyō Sharingan and Hashirama's perfect cells. Obito and Danzō have each done this imperfectly. Obito only possessed one Mangekyō, as did Danzō. Obito was given Hashirama's cells while Danzō incorporated both Shisui and Hashirama's cells in order to attain power, and he hadn't seemed to handle the incorporation nearly as well as Obito. Zetsu can also, ironically, be thought be a Rikudō, as he is quite literally half Hashirama and half Madara. However, this is flawed, because to be a Rikudō is to combine peak Senju and Uchiha powers, and Zetsu is a combination of extremely watered down part of Hashirama and Madara.
While I am listing them first, they are essentially equal to the Uchiha. Yes, Hashirama won their final battle, but he was quite bloodied and it was hinted he has help from his wife to control Kurama, so it's probably safer to say they were truly equals and minor circumstances and luck determined the outcome of the two deadlocked titans. Now, keep in mind that everybody in Konoha that is not an Uchiha is, in fact, Senju. But just like many Uchiha never awakened the Sharingan, so too did many Senju never awaken their birthrights of powerful bodies. And as a matter of fact, doing so was becoming increasinly rare, which is why the Uchiha clan was revered as the strongest in Konoha several decades after the unification of the clans. The Uchiha continued to have ninja awaken powerful eyes and potent chakras, but the Senju slowed in producing ninja with powerful bodies. The best modern-day ninja that most upheld Hashirama's legacy are Naruto, Kushina, and Jiraiya, as they were blessed with special endurance and fortitude, and as such, they alone could channel into senjutsu, or the ability to manipulate nature, which is by no coincidence related to Hashirama's Mokuton, which revolves around spawning natural life. And yes, Naruto and Kushina are Uzamaki, but I'm beginning to suspect that the Uzamaki are branching family of Senju, as Nagato, Naruto, Kushina, and Mito possess some of the most powerful life forces in the series, capable of quelching even Kurama and serving as a host to Madara's eyes, which is incredible impressive when you consider that Kakashi, even now, needs to cover Obito's far less-developed eye for fear of it draining him to death.
The Uchiha possess the other half of the inheritance from the Rikudō, the true god and creator of all ninja, and contrary to common belief, the Uchiha are not a family, but a coalition of families, just like the Senju clan is a coalition of families that include the Akamichi, the Aburama, etc. Only elite Uchiha awaken the Sharingan, and they must develop even more to awaken the Mangekyō. The type of Mangekyō abilities they awaken, however, are dependent on what I suspect to be families. Incidentally, I believe that Sasuke and Itachi are direct descendents of Madara, as Obito said that Susano'o was a rare ability, even among Mangekyō users. To add to this, the jutsu Tsukuyomi must be possessed by Madara to complete his Moons Eye Plan, and the only other Mangekyō user to have Tsukuyomi is Itachi. Obito and Shisui, on the other hand, seem to have possessed the same ability in each eye. Then again, if Sasuke's eyes have a sort of synchronicity, whereas Itachi traded synchronicity for versatility, and I suspect Madara does as well (most likely possessing Tsukuyomi and Amaterasu as well.) In any case, just like elite Senju, elite Uchiha have a leg-up on the rest of the world due to being direct descendents of the original Sage, and thus closer to his power.
However, nothing is ever perfect. And there are certain handfuls of anomalies. Orochimaru and Kabuto, for instance, have circumvented there ways closer to the Sage's power. Interestingly enough, they both wanted strong Uchiha bodies, and not Senju bodies? Why? Because look at the evidence. I believe that it's much more efficient for Uchiha to incorporate Senju DNA, as Obito did, then it is for Senju to incorporate Uchiha DNA, as Danzō attempted. Kakashi is, in essence, proof that you can't just pop Uchiha eyes into a Senju and have the Rinnegan develop. There are also several middle-men. For instance, Nagato is not truly a Rikudō, as he was never genetically modified like Madara or Obito to our knowledge. He was simply an extraordinary Senju, comparable to Hashirama in life energy, that could handle Madara's eyes. This method of ranking also denies intelligence. The name that comes to mind most easily with that statement is, of course, Minato. He was implied to have never mastered senjutsu, and thus not inherited Hashirama's power to the degree that Naruto did. I suspect that it was a stamina issue, as Orochimaru and Jiraiya, believe it or not, clearly delineate the shortcomings in achieving senjutsu. Orochimaru lacked the body, but possessed the chakra-molding ability, and Jiriaya possessed the body, but lacked the chakra-molding ability. Minato, being a genius, has more in common with Orochimaru. Similarly, Itachi and Kakashi also appear to be especially gifted in intellect and talent, so summing their power as an apex of what an Uchiha can accomplish is not quite sufficient, particularly if legendary items are included. The final anomaly are the Jinton users, more specifically Mū. Kekkei genkai were supposedly only inheritable through blood, but Mū somehow trained himself into using a kekkei tōta. That is a special sort of genius that, like Kabuto, Orochimaru, Kakashi, or Minato, may surpass lacking the peak of Rikudō inheritance. But for everybody else, the Uchiha and Senju elite stand atop a precipice above the rest of the world by virtue of their blood alone.
