Sasuke, Suigetsu, Juugo, Karin and Itachi are not members from Akatsuki, they are konoha members who don't know Nagato at all. And Tsunade gave them a mission to assassinate the Akatsuki leader(Nagato). Nagato is aware of that and is waiting for them in the cave here:

They'll be exhausted from their fights U_U taka aside from sasuke are fodder so they won't make that much of a difference..so it'll be exhausted sasuke and itachi vs fully rested pain/nagato. Pain/nagato win both scenarios mid difficulty.

Itachi didn't have knowledge on that technique but he was able to see how it worked and he told naruto and bee what to do in order to destroy it
