both tobi and madara know who they are. also tobi knows all about the history of the uchiha and is bound to know about izanami. madara was the leader if the uchiha clan and read the clan which contains all the secrets of the uchiha so he would know about izanami as well.

remember peolple the izanami is said to help people to find their true self like kabuto for example who wants to be someone else but madara is his self he is not pretending so i dont think izanami would work on him and tobi could kill itachi before he gets a chance to use it
