Pretty sure kisame doesnt know, tobi had him decieved into thinking he is madara from when he was controlling yagura and from then kisame knows him as madara.

I don't recall Kisame adressing him as Madara once, and he did bite off his tongue to prevent Aoba ( i think it was him) to see Tobi's face when he was already going around as "Madara" meaning he was trying to hide his true identity, thus he probably knew who it was

When Tobi revealed himself to Kisame he said who are you and Tobi lied and said Madara Uchiha. But on that chapter he doesnt take off his mask but when Tobi opens his orange mask to Kisame to tell him its not stupid Tobi Kisame recognised him so i dont know you could be right, we have to wait and see if kishi ever explains it.
