The Procedures
Alright, as we all know, Yamato, White Zetsu, Danzo and Edo Madara all have one thing in common; they're all (or were) utilising Hashirama's DNA, one way or another.
Orochimaru used certain type of dna fusion/cloning in Yamato which was different to the procedure he used for Danzo.
With Yamato's experiment, the DNA (Hashirama's cells) was inserted into a young Yamato and used molecular cloning, i.e. allowing Hashi's cells to grow along with Yamato's (Because of the strength of Hashirama's chakra all test subjects died during the procedure, all except Yamato).
Because Orochimaru didn't think this cloning type was successful, he reverted to a different type where instead of using a living human as a host for the procedure, he grew Hashiarama's DNA in the lab - PCR cloning. This way he was able to use the replicated cells through this cloning method and insert it in a host (Danzo & Madara). Therefore restricting the growth of the inserted cells (Hashirama's DNA) any further. The host only needs to draw chakra from the inserted cells.
Tobi improvised by having one Sharingan eye and one Rinnegan eye.
Madara is a step higher. He has EMS and Rinnegan in both eyes but had to switch back and forth.
The perfect experiment, Yamato's procedure, would produce a Rinnegan eye as background and Sharingan Tomoe on top of it. This is the perfect Fusion and this is what Sasuke may gain. While he has that he'd be using EMS on the Rinnegan - Perhaps so powerful and nasty that the Toad Sage just had to prophesize about it?:scorps:
This is why Kabuto needed Yamato or Zetsu. That Hashirama face on Madara's chest was already produced in the lab. So with Sasuke, we won't see Hashirama's face on his chest or shoulders. It would perfectly grow within his own cells without taking over his body.
This is also the reason why Kabuto said he needed a young, live, Uchiha:
Young because an older person's chakra would have waned (as we know from Hiruzen during his battle with Orochimaru)
Live because Yamato's type experiment needs a living host to replicate Hashirama's dna within itself.
An Uchiha because Kabuto needs the power of the Sharingan. I believe Kabuto is trying to unlock the true, ultimate dojutsu through Sasuke:
