Actually we have found 4 Mokkuton users so far; Harashimaru, Yamato, Danzo and Zetsu. Now we haven't seen Zetsu using Mokkuton or Danzo using it properly even though he had used it once for defense against Sasunoo's attack. So far from this point in the Manga I'd say the best Mokkuton user would obviously go to the First Hokage as Yamato had even said that Harashimaru powers were overwhelming compared to his.

As Yamato has said himself, his abilities with Wood Release can't hold a candle to Hashirama's. So it's pretty obvious that the 1st Hokage is the better user of Mokuton. I assume that his wood is stronger, faster, and he also more than likely can manipulate it to its full potential and has better strategies when using Mokuton. We only saw a glimpse of his abilities when we saw him fighting Hiruzen, and even then what we saw was incredible.

you forgot about madara who i think is probably the best because hes had so much more time to master mokuton then hashirama did

eh, i am manga reader and madara never used wood release, if he did than show me page on which he did


but as some people said already, Scary Yamato wins this, followed by Hashirama and than there is Yamato, than Danzo i guess and than Zetsu
