The chakra from the seal still has to flow through her chakra network when she opens it, and with severed chakra points it would not be able to travel through them. Regeneration would not work here. ( unless some chakra threads remain unharmed )

It flows through but it doesn't change the fact that it's separate from her network, because it's stored and can be extracted at any moment. You can think of it as deposit. FRS would end up destroying her Chakra network, but as soon as Sakura activates the seal, additional Chakra would come and heal her.

Can you explain how can the chakra flow through when her chakra network is literally destroyed? Tsunade herself said not even she can repair the damage caused by FRS.

I'm fully aware of how the yin seal works, the problem is, in order for it to work the user's chakra network has to be undamaged.
You can't withdraw your money from the bank if the ATM is broken or if the random fat lady at the bank dies from a heart attack.

Amaterasu can burn through a him cloak that's not a problem never has been. And Tsukiyomi killed his girlfriend by overloading the brain(by showing her 40+ years of memories in a moment causing a short circuit which is also possible. You have to remember it's Itachi who controls the T/S of Tsukiyomi.
