Could MS be unlocked through genjutsu?

What unlocks MS is the emotional trauma one suffers. It doesn't matter if it is reality or a genjutsu.
Illness can be real or psychosomatic, but the symptoms (effects) can be indistinguishable. All that matters is the genuine belief.

You mean like, witness a nightmarish scenario in a genjutsu? Idk, didn't work for Sasuke though. Even after awakening the Sharingan, watching his clan's slaughter through Tsukuyomi for 24 hours didn't do it.

Yes. Mangekyo Sharingan is all about emotional reaction, this was explained in detail by Tobirama. Theoretically if someone cared enough for a loaf of bread and dropped it on the ground by mistake, they could awaken the Mangekyo Sharingan.

I imagine the reason Sasuke didn't awaken it during the two times Itachi used Tsukuyomi in him was that during the first time he was not yet emotionally mature enough to provoke a strong enough reaction to awaken the Mangekyo Sharingan while the second one he knew was an illusion, and thus it didn't have its full effect on him. That is how I see it anyway.

Also, MS trauma isn't any emotional trauma, it's specifically killing someone very close to you (best friend, brother, etc.) is it not?

I never heard of the loaf of bread analogy before but I get it. It's being personally responsible for the trauma you're suffering I guess...

The emotional trauma you go through from losing a loved one is what awakens MS. So yes it is emotional trauma.
