Snake sm is different from frog sm. You have to survive the bite and then you are bestowed the power and right to train in senjutsu.

Juugos original Curse seal comes genetically from his clan but the origin is from the White Snake.

Sasuke can assimilate Senjutsu chakra and possibly unlock Sage Mode, but not in a long run. This is because the key ingredient to attain Sage Mode is siphoning off Nature Energy and balancing it in equal rations with your physical and spiritual chakra. This process would grant the user access to Sage Mode indefinitely. While users who can only assimilate Senjutsu chakra, would attained an incomplete Sage Mode that's likely to be deactivated as soon as their Senjutsu chakra is extinguished and with no chance of re-activating it with reasons that they can't siphon Nature Energy.

Madara absorbed Hashirama's senjutsu chakra and it felt like nothing to him, Madara and Obito were juubi jins and the juubi is composed of natural energy so I don't see what's stopping Sasuke from being able to learn sage mode.

Ryuchi cave is no different than Myoboku and Shikkotsu forest. CM is Juugo's senjutsu chakra and Orochimaru's chakra. Sage transformation also isn't SM if it was then Juugo wouldn't be so weak despite having it.

It was because of Sharingan/Rinnegan and Sasuke had a body for SM.

Regardless of Sasuke's compatibility with Juugo's cells, he has both the Chakra Control and Chakra reserves required to obtain a perfect SM. It could be argued that with Sasuke's CC he could take SM to levels even Hashirama couldn't dream of.
