No. They're comprised of all elements, And are only formed by Rikudo senjutsu users. Bijuu aren't part of it, since The sage & his brother had TSB before the sage himself even invented the bijuu. Also Toneri had TSB without the bijuu.

Sasuke doesn't have all nature transformations,even though the rinnegan grants it and he can't use yin,yang,yin-yang release only six paths senjutsu and kaguya can use it and if he had those he still wouldn't have the understanding of chakra that would be required to make them and he's not otsutsuki.

I think you need all tailed beasts together to make them every time someone made one they had all bijuus but ham
