It a one hit KO depending on if you are a Juubi Jin or not. There is no way someone can use the Night Moth more than once, it puts too much stress on the body. Even before he resorted to use the Night Moth against Madara, Gai retorted he broke a few ribs while using EE or Hirudora so no I doubt you can use it more than once even if you have the necessary chakra.

Are you asking if it's possible for someone to decide to not use any of the other 8th gate techs and instead only use night moth as many time s as they could? If so I'm not sure, I think madara could tell it was guys last attack because he only had enough chakra left for one more move. So I suppose if guy had more chakra he could've used it more than once in theory, but in practice, it probably requires to much pychsical strain

But if the user does not die from 8th gate (like Guy now) then yes it is not as long as the user isn't a ***** and can handle the pain.

BTW I believe that Guy isn't completely useless even though he's now wheelchair bound. All he has to do is open up some gate and he'll be back to normal (gates make the user exceed limitations) and now that there's that lock on his heart, he could use the 8th gates as many times as he wants (isn't that cool?)
