Since he has lived all his life in danger being alone and among threats, he has to be aware and conscious about any sort of risk, considering even the faintest possibility that could lead to danger. Sasuke is naturally suspicious and aware about his surroundings like anyone would be in his situation.

Also, he could have a real reason to do that. Since he considering her to be on their side, he was confused, surprised and angered by the sudden emergence of so many Uchihas once considered to be extinct. He was already disturbed ever since he fought Shin, unable to understand what was happening. And considering it to be problematic, he planned to dispose of them as soon as possible.

Maybe, or rather he didn't expect her to be here at this point in time all by herself. And that sort of thing wouldn't naturally cross your mind immediately considering that he's never seen her, so Sasuke's reaction is justified.

In Sasuke's defense, the possibility of him meeting his daughter so suddenly in a remote location that maybe only a handful of people know about is really slim. Even if he does know about his daughter, taking into account he just faced a youth with a 3t sharingan this kind of reaction is predictable but still shocking.
