I didn't want to assume that her horns were made up of the properties as her actual weaponized bones so I'm asking if Sakura or Tsunade can break the actual weaponized bone Shikotsumyaku.

Then there's no clear answer for your question. Logically, her bones should be on another level when compared to Kimimaro. Canonically, though, they have zero durability feats. Obviously Sakura would die if she tried to punch the All-Killing Ash Bone, but whether or not she'll be able to break it is unclear.

A better question would be if she could punch through Kaguya's hair in its defense mode.

Not sure what you're asking. They can break kimimaro (defense stronger than steel?), however if they get pierced by kaguya's bone they die.

Kimimaro's? Yes, with ease considering she can deal moderate damage to Susanoo.
