The issue here is Kisame getting enough time to create his water dome when he's fighting 3 shinobi. Think he's gonna actually fend off 3 of the best taijutsu users, one having sharingan precog? Itachi already warned Kisame that if he tried to fight part 1 Kakashi that he would get hurt. War-arc Kakashi(I'm assuming is the Kakashi this is using) is leagues above that.

If by some miracle of Jesus himself coming down and buying Kisame enough time to pull off water dome, they can summon Katsuyu and break the waterdome apart/have her get them out of there.

And anyway, if we want to go off of your assumption that he pulls this off, and that Katsuyu can't, for whatever reason you've conjured in your head can't break it, Kisame can only go after 1 opponent in the dome. What's he gonna do with the other two? And he better hope he doesn't **** up and waste his kill on Tsunade or Sakura, who can heal themselves or have katsuyu heal them.

I mean, you're gonna have a hell of a time alone explaining how and why he actually pulls this off, but how he kills all 3 in it will be an even greater thing to watch you try and explain.

The dome moves with Kisame. They have no chance of out running it. Again, katsuya is irrelevant here. Also Kisame in samehada mode took Hachibi's chakra pretty fast. Tsunade and Sakura are nothing compared to that.

Ah, no it reallly is awful. Someone who couldn't even react to Omoi's kick from about half a meter away is not gonna do jack to Kisame, who fight Omoi's master one-on-one. Moreover, she won't even be able to use Taijutsu in the water dome.

Her seal's chakra would get eaten up only if it's opened and samehada makes contact. Sakura and Tsunade aren't dummies, they'll know that the weapon drains chakra so they'll manage to heal their wounds using creation rebirth which isn't unconscious isnstant regeneration.
