Yin Seal Release sends the stored chakra throughout the body. Zero evidence that it stops and disrupts the chakra flow, which is what you need to break Genjutsu.

After focusing and storing an immense of chakra over years/weeks/ months in the yin seal Tsunade can release the stored chakra by dispersing the seal and pumping the stored chakra back into her body. The Yin seal is directly connected to her chakra network, or else she wouldn't be able to use that chakra. So the fact that the seal forcefully pumps back the chakra in her body means it disrupts the flow of her chakra.
