The thing you fail to understand is that even if Tsunade escapes the Dome, it will still be there surrounding Kisame.

3: Tsunade hides in Katsuyu while she splits herself.
4: Tsunade (inside Katsuyu clone) swims to the top/surface of the Water Dome
5: When she reaches the surface she exits and stands on top of the dome like regular shinobi stand on water.

Its countered effectively just like that. If Kisame swims upwards to her he still needs to come out of the dome to attack her because she is on top on the dome outside of it.

But in this particular case Water Dome won't fail. Actually, it's the exact opposite. It will force Tsunade to retreat, using her chakra to summon Katsuyu and make her split into little fodders. And all the little Katsuyu and Tsunade will have their chakra drained by Water Dome simultaneously.

Except the dome itself does not drain chakra. Kisame has to make contact with the target to drain anything and it will be difficult with 100 Katsuyus swimming in all directions.

I don't think 100 katsuyu's is doing anything to 100 sharks. And there's no reason kisame has to chase her down when his sharks can just drag her back.

Either way nothing is stopping him from doing water dome without fusing with his sword.

And it still goes back to my previous point. She escapes then stands on the dome. He needs to come out an attack her or stay i side and just shoot water sharks at her, all of which are tanked.

But she won't be damaging him tho. And there's no guarantee that she can get out via katsuyu method. U said 100 katsuyu's well for each katsuyu there'd be 10 sharks, that without a doubt move faster than her. She gets to the top then the sharks just drag her back down 4 kisame to absorb her chakra.
