Isshin was stronger than Aizen at that time, he didn't last against Aizen.
Unohana on the other hand, said that she is not capable of fighting with him and didn't do anything after Aizen 'defeat' Yamamoto.

What I wanted to say is that Yama even without his zanpakuto, was sure that he can take down Aizen, he could be wrong, but I don't think why would he, he just a moment ago feelt Aizen's reiatsu, he watched whole Shinigami vs Aizen, even though he show all that he wasn't impressed at all.
What scares me is that we are not sure did he refer to Shikai Aizen or Bankai Aizen. If he refered just to Shikai Aizen, there is still possibility (Yama without zanpakuto) for him to beat Aizen, it just depends on how much is he stronger than Shikai Aizen, if he thought about Bankai Aizen, than he win this easily even without his zanpakuto.

Adding his zanpakuto would be overkill, since just his shikai, by Aizen's own admice may be strong to beat him, we again don't know did Aizen refered to his Bankai or just Shikai, I suppose he was referring to his bankai...
