Tobirama was merely distinguishing from her chakra that she is a part of the Uzumaki clan. I would assume having strong life force, etc and knowing Mito he would tell. But, her manifesting the Chakra chains to me signifies that she has similar chakra to Kushina thus could probably hold Kurama.

Her chakra have strong life force no doubt about it ,however hosting the kyubi is something else.

Kushina said all uzumakis blessed with that long live chakra .and kabuto also said all uzumaki have strong life force ,but hosting the kyubi was different because they brought kushina out of all the uzumakis and she was considered special among them to be kyubi host .

So not all uzumakis can host the kyubi ,but they still have thier own chakra like how tobirama could sense it.
