I'm sure edo do have hearts. Why wouldn't they? They have limbs, heads, why should they lack any organs at all?

As for blood, same story, they probably do have it. Summonings should be the evidence enough. Again, if their body is restored, why wouldn't their fluids?

Almost certainly edo have blood and heart.
However, what Madara said most likely means that this blood is still - it's not being pumped by heart and that's what opening the gates is about.

I wouldn't be so sure.
Edos seem to be composed of ground that surrounds them when they are created. It only takes form of paper, but it's not.
Cells are probably formed just like whole edo bodies are, since soil should contain the necessary elements to form them.

I'm not a biologist, though, so if someone knows better, please correct me.
