i dont think so, It was made from the gedo mazo. but madara "injected" his will into it with yin & yang maybe...

I think Obito used kamui into Madara and grabbed the tailed beast possibly Kurama. Then with remaining sage chakra he already possessed it allowed him to absorb more sage chakra and that's how the staff formed,
