quite simple really

ZETSU

the original zetsu that madara created, the one sasuke killed after first using his ems,
somehow managed to survive.

now for why it is zetsu.

the jutsu the 1st hokage gave to sasuke was a jutsu that stops madaras influenc over white zetsu
releasing madaras will from zetsu.

when sasuke jumped from the hawk to attack madara the jutsu was in effect with his esm activated he threw his swords at madara and missed. (or what the reader thought happend) when in all reality the sword was aiming for a white zetsu underground ,thus sending the 1st hokages virus jutsu into all the connecting zetsu .

sasuke didnt mean to get killed by madara, however this happend (not fully dead yet)

if you remeber right before madara seals the bijuu into the gedo mazou he telll white zetsu how he's just a experiment gone wrong, notice the look on zetsu's face he looks hurt by this comment further breaking madaras control over him.

the last step in bbreaking his control is tapping into yamatos fealing which will overide madaras.

now we have the original white zetsu who no longer takes madaras orders wanting to kill him
so decides to help sasuceske .

the original zetsu chakara has'nt been senced by anybody thats why karin is so shocked at his chakara

this is where it gets farfetched, remember the 1st and 2nds other brother?

Suspense is there to lead up to a climax, a mere Zetsu contradicts that by being anti-climatic.

Sakura and Shizune done DNA tests on Zetsu and the found out that
the DNA resembles the first hokage Hashirama.

The black part of zetsu is because of Madara's will.

Itama has nothing to do with Zetsu.

However I like the part of the theory, where Hashirama's Chakra is meant to stop
Madara's control over Zetsu. I would like to say, stop Madara's control over Hashirama's cells.
Maybe its one of the reasons why Madara spit out the guard and jar, because he is starting
to lose control over the cells.

yeas they did atleast looks like they did soul transfer is a possibilty or a unknown kinjutsu that took hisw senju dna to use

madara stated someones ealse in the uchiha tried to add senju and uchiha dna together

that is a really good theory however I think it is SHISUI UCHIHA now i have nothing to really back this theory but think about this he is the only character who is left unknown itachi was the last one to see him he was never confirmed to be dead and it would be fitting for him to help his friend (itachi's) little brother
