Uchiha loosing/weakening their visual powers by using them is a reoccuring theme throughout the manga. The most powerfull visual jutsu of the uchiha that we know of should be the infinite tsukoyomi and I think its save to assume that this jutsu as well has its cost...

Afterall, he can use rinne tensei on Sasuke without dying since it would be immediate and only one person. Also, he can read minds. 1.Madara could be talking about Obito`s rinnegan.....he isnt sure if he will get it or not(minato and kakashi might stop the black zetsu)
2.Madara could be talking about sasuke`s EMS in a metaphorical way(a possibility of gaining sasuke`s allegiance and another eye to transform into a rinnegan.....but he isnt sure about it and sees immediately after that sasuke wouldnt cooperate so he stabs him....which clarifies his words that the eye wouldnt stick with him
3.Madara could be talking about the ultimate eye of the juubi? the one that is both rinnegan and sharingan........

Chapter 449, second page, top center panel (yeah you gotta go look for it like I did) Flower brat said "If he uses it now, with these chakra levels..." and "...yet he still wants to follow this child". Decypher that. The justu is a known jutsu so it must've been used before. As for mind-reading, I looked back and, though I thought obito said in his mind that he could go protect his friends now that he has awakened his sharingan, he actually said it aloud and Madara responded.
