I've been wondering this.
So I did some research. Kisame himself said he was no match for him. Itachi also stated a best case scenario would be for both of them to die, killing him as well.
My conclusion: Jiraya would have soloed them....here's so facts:
Kisame
Pretty much any water attacks will be countered by Road oil flame bullets. This sage jutsu is a mixture of wind,fire and toad oil....which is pretty combustible like grease. What happens when water hits a grease fire?
He also can make a lake of burning hot toad oil with this attack as he did to pein.
Taijutsu vs taijutsu Kisame loses. Jiraya is to fast in sage mode and hit one of peins bodies hard enough to knock him out 1 time.
He can also create a expanding barrier to detect attacks in a radius much like he did vs prob.
It it is my belief that jirayas genjutsu would destroy kisame.
Itachi
As far as base fire jutsus, jiraya has the upper hand since roads can spit oil increasing the power of the flames.
Itachis best bet would be tsukyomi but since ma and pa frogs are needed to go into sage mode any genjutsu is rendered useless.
As for amateratsu, jiraya would need a 2 step plan to counter it. First, he would need to cover himself in his hair with needle jizo, then use the sage art: hair sonbon to shoot off the flaming pieces. And again, his barrier hirsute would help him detect this attack.
If things get tough, jiraya still has 2 ways to win this fight.
1. Sage art: frog song....I do not believe kisame or itachi can counter this. Yes it's genjutsu and itachi is the master of genjutsu but
there are 2 things to take into account here. 1. This IS NOT a visual genjutsu. 2. We have learned that sage techs don't work like anything else.
2. Reverse summoning toad gourd. All he has to do is reverse summon them one at a time inside the aciditic belly of this frog....in which he has the advantage.
