I've seen some interesting theories floating around on other fourms that the Uzumaki clan weren't just killed off by the other villages but were really killed as a part of Madara's plan. Now the counter argument to this is how was Madara able to move around when he was probably old by this time but lets remember this:

The Mist was already known as 'the bloody mist' before Obito controlled the Mizukage which I think is Madara's doing. Now what would Madara have to gain from this? Well think about it, the Uzumakis were known for their sealing techniques and being feared for it by other villages they were destroyed. Now I think that the destruction of the Whirlpool village was committed by the Mist Village. They had the Sanbi at the time which was their largest and most dangerous war potential so the Whirlpool Village, being allies of the Leaf would have sealed the Sanbi away to protect Konoha. Madara as Mizukage could have convinced the Mist higher ups that the Uzumakis were a danger to the Mist village by sealing the Sanbi when in fact he was really more concerned about them sealing the Juubi in the future thus destroying all hope of achieving Tsuki no me.

Nice theory well made, bu answer this: How do you explain the fact that a clan that was powerful like the Uzumakis was kiled off easily while thy had allies like Senju? Well I'm confident that iUzumkis were defeated when it was during he 1st shinobi was, afterall we not know anything about it.

i dont think it was the 1st ninja war, possibly the 2nd great war seeing how kushina arrived to konoha when she was a kid and minato only took part in the 3rd great war

If this was during war time, then it's quite likely that the Leaf were occupied with fighting against the Rock and Cloud ninja. Also if you look at this map:

Unlike the other hidden villages, The Mist had a clear path to sail and invade Uzushiogakure from their location.

The Uzumaki clan were exterminated because they were the perfect candidates to seal bijuu and become the perfect jinchuuriki for them too. Since they had the perfect life force, the bijuu wouldn't tear them apart from the inside.

The Wirlpool village (if they were still alive) would be the most dominant village, with all their sealing techniques and their perfect candidates for jinchuuriki.

But he still seemed able to used the Mist village to his advantage which came after the destruction of the Uzumaki clan.
