Considering that Gai is much stronger and faster than Lee, and considering that he can easily open up to 7 gates that further increase his power and speed, Tsunade, politely saying, is at a serious disadvantage

Tsunade also can increase her speed with chakra control.

Reaction time doesn't help you if your body simply cannot provide you with enough speed to take an advantage out of it. Base Tsunade is 1.5 ranks slower than base Gai. Gated Guy is out of her league.
Also, I don't know who told you thank chakra control can imbue your speed. For your information, chakra control lets you waste LESS CHAKRA while using jutsu and also allows you to focus chakra in some precise points. Tsunade hasn't shown any movement speed increasing jutsu so chakra control doesn't help her in this area at all.
Also, Tsunade hasn't presented any advanced taijutsu feats so far. Any fodder could could punch some holes if they had enormous strength.
