Madara has a vast amount of knowledge.....after his apparent 'death' at the hands of hashirama he spent years in his secret hideout and gathered a lot of knowledge about the juubi and the sage probably....because the moon's eye plan was originated by him, he awakened the rinnegan and it was him who first unsealed the gedo mazou from the moon. So what i think is that the trump card he was talking about has to be something which only he and the black zetsu is aware of. It can be anything....though my assumption is that it may be a jutsu that enables the user to take control of a tailed beast inside a jinchuuriki. That can only be possible if the jinchuuriki himself has very low resistance to the external control. Obito probably is not in complete control of the juubi inside him which can give madara the advantage at the moment. And why he needs to hurry? Because if somehow obito gains control of the juubi soon....then things will get difficult for madara to carry out his plans.....

I dont think so.
Madara stated its his trump card, so he had it in reserve all the time.
Obito becoming the Jinchuriki was unexpected, he cant have a trump card for an event that he didnt expect and didnt know of until the very moment it happened.

His trump card has to be something else, something that can counter Obito without specifically beeing created for that purpose.
