Does Itachi knew tobi was a Uchiha With Mangekyō Sharingan?

When Uchiha Itachi wiped out his clan he let his younger brother know that the ultimate nature of their clans bloodline power was the Mangekyō eye.
Itachi told Sasuke that only three people had handled the Mangekyō: Uchiha Itachi, Uchiha Shisui and some unknown third individual.
Now the third must be Madara Uchiha.Also he cant mistake Tobi with Madara as Madara got Eternal Mangekyō Sharingan.

Also he wasnt in the the top 3 guys 2 activate Mangekyō Sharingan.
Madara, Izuna, Obito+Kakashi and Shisui had handled/activated MS before him(except kakashi).

So does he knew that Tobi had Mangekyō Sharingan?

This. There is no defined way to tell an an EMS from a MS unless you see both eyes separate before they're fused. Regardless, Itachi believed Obito to be Madara so yes he knew of the MS.

^ what this guy said. Itachi believed Tobi to be Madara, and he planted that hint so that Sasuke would grow up distrusting Tobi I suppose (as shown in their fight when Sasuke resolved to kill Tobi after Itachi). of course, Itachi's plans with Tobi all flopped in the end

Ok all u guys who have posted above one question before we used 2 know Tobi's identity he could be any random shinobi who might have stolen any Uchiha's eyes or any other Uchiha himself plus we can never know whether Tobi showed his Kamui 2 Itachi.so how can u say Itachi thought he was Madara?
but maybe u guys r right in a way that he definately have assumed that Tobi is Madara rather than any other Uchiha or Shinobi.
